How plausible would a successful American uprising be in the novel Man in the high castle

It would be pretty difficult but if rebels could sieze the missile silos in North America at the beginning they could possibly have a chance.

Considering Japan and Germany are about to wipe each other out, I'd say that once the German and Japanese Home Countries are glowing, it'll be pretty much a given.

According to the map in the war room scene Berlin built a line of silos on the edge of the Neutral Zone aimed at the Japanese. If the range was great enough to strike the Reich proper they would be a good deterrent. One irony is that the second season shows that the citizens of the Greater Nazi Reich are far more hardline in their "traditional values" than the citizens of the Grossdeutsch Reich.
