What if Spain never joined in on the Seven Years' War?
I'd imagine the only way to do this is to not have the British attack Manila and Havana.
With Spain out of the picture, I'd imagine Portugal also would not join the war.

What would happen to Louisiana? Considering that the territory was ceded to Spain, I'd imagine that the British would attempt to claim it.
If they did win it over, how would it effect the American Revolution? Would we see the Americans attempt to win it like they tried to do with Quebec?
Would the city of St. Louis ever be made? Considering it was made following the war, while Spain had held it.

How would this effect fighting in Europe? Would it at all?

UK held Minorca and Gibraltar and those two factors would be hard for Spain not to join. You need U.K. To have taken Ceuta instead of Gibraltar for Spain not to be in war.

Spain got Minorca back but were unable to get Gibraltar back in 7 year war and have been trying to get it back ever since.
 
Spain joined the war in 1761 when the Bourbon Family Compact was signed. Britain then attacked the Spanish colonies - it was not the other way around.

France ceded Louisiana to Spain in 1762 as compensation for the British occupation of its colonies. This was a secret treaty; the 1763 peace treaty actually states that Louisiana is still French, with the Mississippi River being the dividing line between the French and British territories. I would guess that it would simply remain French after the war. (It's conceivable that Britain would have attacked Louisiana in an alt-war where there is no Spanish entry, but it doesn't seem to have been a priority for them.)
