On multiple occasions, Mithridates VI of Pontus offered to make an anti-Roman alliance with the Parthian empire, but the Parthians never accepted.

What if they had? Could a Pontic-Parthian alliance have checked Roman expansion?

At this point, Rome was no threat to Parthia. Pontus (or at least Armenia) on the other hand were much closer to home, and much more capable of meddling with the internal affairs of Parthia (especially given that the Mithridatic family claimed descent from the Achaemenids), so they were natural enemies
