@SpaceOddity's got a similar situation in his TL "Now Blooms the Tudor Rose", but I thought it would be fun. Of course, this necessitates James V to live a bit longer than a mere six days after Mary's born (although I suppose Mary's sister could be a twin), probably long enough to get his wife preggers again. Would both girls be sent to France during the Rough Wooing (which may or may not take place here, if James V is a bit longer lived (he still dies and leaves Mary to a pretty long regency), or they consent to the younger one being married to the prince of Wales? Would both marry French princes (one to the dauphin? and one to some other prince du sang I'm not sure who would be available, since Charles IX was pretty young for Mary, and would still be if her sister is 12-18mos younger than she is.

So what happens to Princess Margaret (figure much as James may have disliked his mom, naming the girl after James' mother who shares a name with Scotland's royal saint is most likely)? Future queen of England to Edward VI? Queen of France to François II instead of her sister (in which case what happens to the wee queen)?
