Sons of Francis I die in captivity in Spain

King Francis I of France, after being captured by Spanish/Imperial forces at battle of Pavia, had to send his two older sons to Spain to regain his freedom according to Treaty of Madrid. What would happen if sons of French King died not long after arriving to Spain, say they fell ill, so Charles V send them doctors-Spanish doctors at the time were able to kill even healthiest person, so it is easy to imagine that something went wrong.
What happens next? Charles V lost his hostages and would be blamed for their deaths, Francis would hate Habsburgs even more on one hand, OTOH he does not need to care about his sons in Spanish captivity anymore. Charles V would not enforce match between his sister and French King, while Francis, after loss of two sons, would seek for new wife, there is only his youngest son Charles left, so Valois line is in danger.

Francis just got a even bigger stick to beat Charles with. And the death of the princes might even cause him to loose support in Europe.

Now the question is who Francis would remarry instead of Eleanor? Could he marry his sister in law, Renée?

Charles V is going to have really bad press now. In addition to Sack of Rome he is accused of murder of princes.

Francis could marry Hedwig Jagiellon-Hedwig's uncle, Janos Zapolya, was Habsburgs' enemy, anti-King of Hungary, supported by France and Ottomans. Janos came with idea of marriage of his niece with French King. Alliance with Zapolya and Jagiellons would be useful for Francis. IOTL Francis preffered Hedwig's half sister Isabella, due to her Sforza blood and claims to Milan, but she was 6 years younger than Hedwig and Francis hardly could wait so long with remarriage.
What is going to happen with Catherine di Medici BTW?

AFAIK Francis repudiated his concessions almost as soon as he was back to France even while his sons were in Spain: it was quite obvious that Charles is not going to kill them.

As for the hate, I’m not sure how much these two had been hating each other on a personal level and how much it was “just business, nothing personal”. In practical terms, it was not a matter of hate but rather of a possibility to raise more money and troops to continue the war. Francis did not give up in OTL and would not in your AH and the issue of the hostages and a marriage would change little. An issue of the succession is, however, serious because Charles of Orleans died before Francis. Who was the next in line?

It is interesting how the changed situation would be reflected in the Treaty of Crepy (as I understand, by 1544 Charles was running out of money and had to deal with the HRE problems) without a convenient OTL option of having Charles of Orleans marrying one of the archduchesses with the seemingly convenient territorial rearrangements.

Circumstances of the death of Charles d'Orleans tell us, that he certainly was not the most intelligent prince of House of Valois, but OTOH it could be easily butterflied away. Francis still have time to sire more sons-POD happened 20 years before his death.

Sorry, but how many intelligent ones had been there post-Louis XI?

One issue is if Charles is destined for an early, idiotic death, is that Brittany will leave the French main line, going first to Madeleine and then (provided she meets a similar early, childless death ala OTL) her sister Margaret. Unless Madeleine/Margaret leave a female heiress there's no chance of Brittany returning to France besides a Ptolemy-esque marriage between Madeleine/Margaret and her younger half-brother, which ain't happening.
