He has never been so good.

Harold McMillan.
From Wikipedia:

The Profumo affair may have exacerbated Macmillan's ill-health. He was taken ill on the eve of the Conservative Party conference, diagnosed incorrectly with inoperable prostate cancer. Consequently, he resigned on 18 October 1963.

But what if Profumo affair had never came out,and the diagnosis for McMillan had been correct?

Yes, I would say it's possible. Alec Douglas-Home came startlingly close to a victory in OTL to say A) the Tories had been in power 13 years, B) he was considered out of touch and old-fashioned, and C) Profumo.

I'm not sure why Labour weren't in a stronger position, I think it's because Gaitskell died in 1963 and they had had a messy punch-up over the leadership election that eventually ended with Wilson's victory.

A No Profumo/No Misdiagnosis TL would be a gripping read. I wish I had anything like the knowledge required to have a bash.
