What if Black Prince was widowed not long after birth of Richard II and marries Constance of Castile, OTL duchess of Lancaster as his second wife (John of Gaunt likely marries her sister Isabel instead). Say that Black Prince has at leas one child with her. What would be effects? Most obvious one-John of Gaunt's Iberian adventure is butterflied away-his brother's wife has better claim to Castile ITTL than his own wife. As Gaunt's attempts to get Castilian throne failed anyway, it would be good thing for him-he would not waste resources for such fruitless campaign. Balck Prince's half Castilian child OTOH would be too young to be serious claimant. That means also that Anglo-Portuguese alliance is weaker than IOTL without Gaunt's involvement.

This child also butterflies any Mortimer claim to English throne, whether it's male or female. Male might butterfly Henry IV's usurpation, as Parliament would not tolerate setting aside king's brother in favour of king's cousin.

There would be massive butterflies, existence of said child (let's say that it is a boy, Edward, Duke of Clarence, b. 1372) may prevent Richard's downfall in the future.

There would be, but I'm not sure if Richard's downfall would be affected. And what would be Clarence's personality? Because if he was similar to another Clarence (George of Clarence, to be exact) in personality, than TTL Richard's reign would look....interesting.

If he is going to resemble another Clarence then better if said Clarence is his uncle Lionel.

Not much is even known about Lionel's personality, apart from him being easily bored and being unusually tall and athletic (but that's not a personality).
Still, he could not be worse than George.

Depending on EoC personality and relations with brother, he could be the one who'll rebel against Richard.

It's hard to be worse than George, but yes, EoC could rebel against his brother (maybe joining lords appelants) and he even might be dead before Richard, but who could duke of Clarence be married to?
