Japan's eagerness to attract Jews was based on the popular belief that Jews were wealthy and had a lot of political influence, but only a small portion of Jews went to Manchuria, and many of those that did eventually moved into Shanghai.

Challenge: Have more Jews immigrate and settle in Japanese Manchukuo in such a way that Japan gains extreme favor from the Jewish population.

What would happen, especially to Militaristic Japan's relationship with anti-Semetic Nazi Germany and WWII?

I honestly don't think anything would happen here- it was always an alliance of convenience. IOTL some of the Japanese Embassy officials in the Reich (to their eternal credit) really stretched the boundaries of legality to get Jewish refugees exit visas to Japan and it didn't affect relations with the Nazis.

If there's much more Jewish settlement in the Japanese Empire and they become seen as a group favoured by the Imperial administration then there will be consequences. If the general course of history goes as per OTL, (a) the Jewish settlers in Manchukuo are going to be targeted by the Chinese and (b) there may well be somewhat less sympathy for the Jewish people as a whole after the war and may actually fuel some antisemetic propaganda ("While Hitler slaughtered Jews in Europe, Jewish settlers supported the filthy Nips in raping Asia! Just see how the traitorous Jew will slither over to anyone who provides a chance for profit!")
 
If we're talking about an interwar settlement program, such a move would definitely alienate the anti-semitic White Russian mercenaries and organized criminal elements in Manchuria that the Japanese relied upon to foment internal dissent across the border in Soviet territory.
