I would argue that if there was no American colonisation, which can be achieved by the Tainos killing or capturing Columbus without him coming back, thus cancelling Spanish voyages into the deep Atlantic, Spain doesn't become a superpower/hyperpower and is left only with a very unruly Holy Roman Empire and Italy that are challenged by France - the second most powerful state in Western Europe - which would mean a more successful spread of Protestantism by converted duchies and fiefdoms that invade and overwhelm both Spain and France (who are too busy fighting among themselves to effectively go after the Protestants) and thus forcibly convert them to Protestantism, whether Calvinism or Lutheranism. It's quite possible this would force the Pope to escape to Britain, which would become the bastion of Catholicism in Western Europe.
