A lot of anime would do surprisingly well as musicals. Macross is an obvious example, you just need characters besides Minmay singing, even if this would be a bit out of place given the OTL show. The sequel Macross 7 could easily be reworked to be a musical--just change the lyrics of the songs Fire Bomber is singing a bit. Macross 7 kind of does have musical elements already given when they introduce a specific song in the show (i.e. "My Soul For You", "Holy Lonely Light", "Power to the Dream", etc.) and what's going on in the arc said song is introduced or played frequently. The same can be said for the Symphogear franchise, where every episode has the characters singing songs with lyrics reflecting their past, what they're feeling at the time, or what they wish to happen. Of course, in Macross, music has power over the villains (various aliens) and the main characters are always musicians and the people closest to them, and in Symphogear, the characters are powered up by singing to fight monsters/evil alchemists/whatever.

The main thing would be jumping from shows where the characters are musicians to when they just randomly break out in song. That's what really makes a musical to me IMO, since I don't consider Macross 7 or Symphogear as musicals. A proper Macross musical would have the villains singing too, after all--I'd depict their music as harsh and alien sounding, although given the original Macross was made in 1982, that might be difficult. Maybe use music inspired by dissonant modernist classical with vocals on top--that's plenty "abrasive" and fitting for the Zentraedi compared to the more "human" sounding music of Minmay (other characters like Hikaru, Misa, etc. would also be singing this sort of music). This would incidentally make SDF Macross more difficult to dub as Robotech. The villains in Macross 7 and Frontier could have their songs be aggressive and dissonant EDM music. As for Symphogear, most of the villains sing anyway, but you could have more lighthearted songs (there already are some anyway) and songs the villain sing when they aren't fighting the heroines.

That's just some OTL anime I'd identify as "musical". But you could turn a lot of good anime into musicals. Any Gundam show could work as a musical, as odd as it might be to have, say, Char Aznable (his namesake Charles Aznavour wouldn't be suitable I feel) or any of the Zabi clan singing. Or maybe Fang of the Sun Dougram with its anti-imperialist themes resulting in characters often breaking out into spirited folk songs inspired by Pete Seeger or Victor Jara (and I can only imagine what you could do with a musical take on Armored Troopers VOTOMS from the same director). Or Hokuto no Ken inspired by some straight up metal stuff, with Ken singing some 80s metal song every time he's killing the bad guys ("YOU WA SHOCK!" just isn't enough). The list goes on and on, encompassing any popular anime in the 90s, 00s, and beyond. Another anime classic, Legend of the Galactic Heroes, actually was done as a musical in recent years, although I have no clue if it was any good (and given the Romanticist classical music the show borrowed, this doesn't seem to hard to make a Wagnerian opera out of it).

Overall effect in anime is to make sure that your average seiyuu--male or female--will be expected to also be able to sing well if they want to reach the highest levels of popularity and fame, although to some degree I'd expect some seiyuu would have their character's singing voice done by an actual singer. I could imagine some anison singers like Masaaki Endoh or rock/metal singers like Eizo Sakamoto doing a lot of singing voices in "musical anime", maybe as typically the role of elder characters or villains and such. As a personal wank of my own interests, I'd love to see singers like Yama-B or Fuki overdub some voice actor/actress given how dramatic their voices are.

Basically, it could seriously work if the average anime character burst out in song to the same degree as the average character in a classic Disney movie (an influence on anime). And even better if it was made sure the music matched the characters.
