Sigimund's I first wife, Barbara Zapolya gave birth to two daughters - Hedwig and Anna. Anna lived 5 years and then died. But what if she didn't die so early? Let's say that she inherits her father's lifespan and outlives her brother, dying in 1580. Who would be considered as her husband?
 
Ludwig or Ernst of Bayern? Ludwig was candidate for Hedwig's hand, Ernst was considered for one of Bona's younger daughters. Johann of Küstrin, brother of Hedwig's husband (also considered for her) could be seen as waste (two sisters for two brothers) but also is possible. Dauphin too, Sigismund wanted to marry Hedwig to him, but François preffered Bona's daughter Isabella for his son, due to her claims to Milan. Or François I himself, although he'd rather marry opder Hedwig if he was interested in Jagiellon match.

Johann of Kustrin seems like the most likely candidate. French match would be considered as a threat to Bona's children and Ernst of Bayern had no land on his own for a long time. In addition, Johann of Kustrin having a male heir with a prospects of getting PLC's throne once Sigismund II dies would be the most interesting scenario possible. The division of Brandenburg would continue and there will be no Prussian inheritance for electors of Brandenburg.
 
Another option is the son of Georg the Bearded of Saxony. Yes, it's her first cousin, but the pope granted Karl V and Isabel of Portugal; Isabel's brother, Joao III and Karl's sister, Katharina; Joao's dad and Karl's sister, Eleonore; dispensations, so it's not unthinkable. Especially if the whole Reformation thing threatens as OTL.

Which one? Johann is too old, while Friedrich was retarded (very). So no chance there. If their cousin Severinus of Saxony survived, he'll be perfect candidate.
 
A potential new alliance? Or Mary of Hungary dying prematurely, that would necessary a new regent for The Low Countries.

Can Anna be considered for Charles V after the death of Isabella of Portugal?
No. Charles lost interest in marriage at all after the death of Isabella. He even became a monk. And Saxon marriage seems very unlikely because of reasons which @Jan Olbracht mentioned. The only serious contestor would be Henry of Portugal (backed by Habsburgs).

Could the Congress of Vienna go differently and see Anna married to her cousin (Lajos II) in Hungary?
